No.
Due to the high current (about 670mA) required by the Harmony Singer 2, it cannot be powered from the Pedal Power 2 DC outputs.
However, you can power it from the AC courtesy outlet with its adapter/wall-wart.

The main difference is that Pedal Power 2 Plus outputs 5 and 6 have high current capacity (250mA) in the 9VDC setting. Whereas, these outputs on the Pedal Power 2 (non-Plus) have only standard capacity (100mA) in the 9VDC setting.

Yes, it is possible to power both with the Pedal Power 4 x 4, setup:
To power the HX Effects with Pedal Power 4 x 4, use two 9VDC high current capacity (400mA) outputs with a HX Current Doubler Cable
To power the Ground Control Pro with Pedal Power 4 x 4, use a 9VDC high current capacity (400mA) output with a 2.1mm/2.5mm Reverse Polarity Barrel Cable, part# PPL6, PPL6-R
Also/FYY, the Pedal Power 2 Plus DC outputs (even 5 and 6) cannot be used to sufficiently power the HX Effects but it can instead be powered from the AC courtesy outlet with its adapter/wall-wart.

Yes, Pedal Power X4 can power the Boss ES-8, easily.
To power the ES-8 with X4, use any of the 9VDC outputs with one of the all black (standard polarity) cables.

Yes (the actual current draw of the TC is less than 250mA).
To power the TC-Helicon with Pedal Power 2 Plus, use output 5 or 6, DIP switch in the 12V (away from normal) setting with one of the all black (standard polarity) cables supplied.
To power the Line6 wireless with Pedal Power 2 Plus, use a Current Doubler Adapter, part# PPAP in outputs 4 and 5 (or 6 and 7), DIP switches set normal (9VDC) with one of the all black (standard polarity) cables.

As for damaging a pedal with excess voltage, it can happen quickly, especially if the excess is large but sometimes it can be slow (for example, it might take 15 minutes for some component to over-heat and fail).
Usually, after any such powering error, if the pedal still works (with the proper power connected) it's fine/undamaged.

The trouble that the lead with the white stripe does not work is fairly common but (usually) it is merely a result of how the output splitter is designed (and not a malfunction).
The lead with the white stripe has its ground lifted. The device powered from this lead must get its electrical ground connection via the audio signal path. So, this device will need to be connected to the rest of your pedals before it will power on. This design is intentional as it eliminates the possibility of creating a ground loop (a common source of unwanted hum/buzz when daisy chaining power).
I suspect the Output Splitter Adapter will work properly once both pedals (on the splitter) are also included together (via the instrument cables) as part of the same audio/signal path.
